Naše oddělení se zabývá studiem mitochondrií ve fyziologických a patofyziologických procesech buňky a organismu. Mitochondrie jsou hlavním zdrojem buněčné energie, ATP, potřebné pro chod buňky. Zároveň TÉŽ dochází k produkci kyslíkových radikálů, reaktivních molekul způsobujících oxidační stres. Ten má negativní dopad na funkci buňky samotné. Důsledkem oxidačního stresu dochází často k nevratnému poškození buněčných proteinů, lipidů i samotné mitochondriální/jaderné DNA, nositelky genetické informace, což v nejkrajnějších případech vede ke smrti buňky. Chronický, byť relativně mírnější oxidační stres doprovází řadu patofyziologických onemocnění mezi které patří neurodegenerativní onemocnění, jako jsou Parkinsonova či Alzheimerova choroba, ale např. i cukrovka 2. typu a pulmonární hypertenze, jejichž etiologii studujeme. Nevratně poškozené mitochondrie je nutno z buňky odstranit pomocí specifické dráhy autofagie – mitofagie. Mitofagie je důležitý proces pro udržení kvalitní mitochondriální výbavy a její narušení je doprovázeno řadou onemocnění, jako např. cukrovka 2. typu, kde tento proces studujeme. Mitochondrie jsou semiautonomní organely a mají tedy svoji DNA, mtDNA. Genetické manipulace mtDNA jsou poměrně obtížné a doposud nebyla vyvinuta uspokojivá metoda, která by umožnila např. genový silencing či in situ kvantifikace genetických mutací na mtDNA. O vývin takovéto techniky se snažíme. Jelikož mitochondrie jsou velmi malé organely a rozlišení běžně dostupné světelné i fluorescenční mikroskopie není dostačující pro jejich studium, využíváme techniky speciální vysokorezoluční fluorescenční mikroskopie, jejíž prototyp jsme nedávno zakoupily pro naše oddělení a dále tuto metodiku rozvíjíme pro využití při studiu mitochondriÍ. V neposlední řadě se zabýváme také úlohou rolí mitochondriální signalizace v rakovinných buňkách, kde ačkoliv mitochondriální aktivita je často velmi utlumena, přesto jsou zde specifické enzymatické reakce, dráhy, jejichž studium je důležité pro vývoj budoucích protinádorových léčiv.
V oblasti aplikovaného výzkumu se naše laboratoř zabývá vývinem nových nosičů léčiva, tzv. fotosintizéru, využívaného ve fotodynamické terapii při léčbě zhoubných nádorů. Prvotní výsledek přinesl nález, který byl patentován u Českého úřadu pro patenty a vynálezy.
