Institute of Physiology of the Czech Academy of Sciences, we provide systematic training for students enrolled in bachelor\u2019s, master\u2019s, and doctoral study programs. In this area, we cooperate with a number of Czech universities as well as with renowned foreign institutions. Offers of bachelor\u2019s and master\u2019s thesis topics can be found both on the Institute\u2019s website and in the information systems of the respective faculties, where Institute staff members are registered as supervisors or, where appropriate, as consultants. Scientists of the Institute of Physiology supervise more than 60 undergraduate students. Kv\u016fli vysok\u00e9 homologii ortosterick\u00e9ho vazebn\u00e9ho m\u00edsta mezi jednotliv\u00fdmi podtypy muskarinov\u00fdch receptor\u016f je hled\u00e1n\u00ed selektivn\u00edch ligand\u016f komplikovan\u00e9. Tato diplomov\u00e1 pr\u00e1ce se zam\u011b\u0159\u00ed na farmakologickou anal\u00fdzu muskarinov\u00fdch ligand\u016f nov\u00fdch chemotyp\u016f. Bude obsahovat jak vazebn\u00e9, tak funk\u010dn\u00ed studie aktivace receptoru. Stanov\u00edme afinitu k\u00a0jednotliv\u00fdm podtyp\u016fm muskarinov\u00fdch receptor\u016f, a zhodnot\u00edme vazebnou a funk\u010dn\u00ed selektivitu nov\u00fdch ligand\u016f. C\u00edlen\u00e1 signalizace umo\u017e\u0148uje ovlivn\u011bn\u00ed specifick\u00fdch sign\u00e1ln\u00edch drah aktivovan\u00fdch receptorem, co\u017e vede k&nbsp;efektivn\u011bj\u0161\u00ed l\u00e9\u010db\u011b s&nbsp;men\u0161\u00edmi vedlej\u0161\u00edmi \u00fa\u010dinky. Zam\u011b\u0159\u00edme se na inovativn\u00ed p\u0159\u00edstup vyu\u017e\u00edvaj\u00edc\u00ed chim\u00e9rick\u00e9 G\u03b1 podjednotky. T\u00edm, \u017ee se vym\u011bn\u00ed C-koncov\u00fd segment G\u03b1 podjednotky, lze &#8220;p\u0159esm\u011brovat&#8221; signalizaci jednotliv\u00fdch G protein\u016f ke stejn\u00e9mu efektoru, \u010d\u00edm\u017e se zjednodu\u0161\u00ed m\u011b\u0159en\u00ed a umo\u017en\u00ed p\u0159\u00edm\u00e9 srovn\u00e1n\u00ed \u00fa\u010dinku agonist\u016f. C\u00edlem magistersk\u00e9 diplomov\u00e9 pr\u00e1ce je zjistit, zda tento p\u0159\u00edstup skute\u010dn\u011b zachov\u00e1v\u00e1 specifick\u00e9 preference agonist\u016f pro jednotliv\u00e9 sign\u00e1ln\u00ed dr\u00e1hy. V\u00fdsledky t\u00e9to pr\u00e1ce mohou p\u0159isp\u011bt k&nbsp;v\u00fdvoji spolehliv\u011bj\u0161\u00edch a efektivn\u011bj\u0161\u00edch n\u00e1stroj\u016f pro v\u00fdvoj l\u00e9\u010div c\u00edl\u00edc\u00edch na muskarinov\u00e9 i dal\u0161\u00ed receptory sp\u0159a\u017een\u00e9 s&nbsp;G-proteiny. Ketol\u00e1tky jsou zdrojem energie za podm\u00ednek, jako je hladov\u011bn\u00ed a nebo kalorick\u00e1 restrikce. C\u00edlem pr\u00e1ce je pou\u017e\u00edt metabolick\u00e9 mapov\u00e1n\u00ed (stabiln\u00ed isotopy uhl\u00edku) ke sledov\u00e1n\u00ed vzniku ketol\u00e1tek a jejich distribuci v&nbsp;modelov\u00e9m organismu my\u0161i s&nbsp;pomoc\u00ed LC-MS. Pro spr\u00e1vn\u011b se\u0159\u00edzen\u00ed pot\u0159ebuj\u00ed informace o sv\u011bteln\u00fdch podm\u00ednk\u00e1ch ve vn\u011bj\u0161\u00edm prost\u0159ed\u00ed, zprost\u0159edkovan\u00e9 pomoc\u00ed centr\u00e1ln\u00edch hodin, a informace o aktu\u00e1ln\u00edm stavu bd\u011blosti \u010di sp\u00e1nku. Modern\u00ed \u017eivotn\u00ed styl naru\u0161uje soulad mezi na\u0161\u00ed aktivn\u00ed a sv\u011btlou \u010d\u00e1st\u00ed dne. Dopad takov\u00fdch podm\u00ednek na hodiny v\u00a0r\u016fzn\u00fdch \u010d\u00e1stech mozku nen\u00ed dosud prozkoum\u00e1n. Navrhujeme jejich nov\u00e9 vyu\u017eit\u00ed jako chronoterapeutik vyu\u017eiteln\u00fdch pro l\u00e9\u010dbu poruch CR zp\u016fsoben\u00fdch genetick\u00fdmi (d\u011bdi\u010dn\u00e9 poruchy sp\u00e1nkov\u00e9 f\u00e1ze, nap\u0159. FASPS) nebo vn\u011bj\u0161\u00edmi (p\u00e1smov\u00e1 nemoc, soci\u00e1ln\u00ed jet lag, pr\u00e1ce na sm\u011bny) faktory. Projekt zahrnuje identifikaci kandid\u00e1tn\u00edch l\u00e1tek skr\u00ednov\u00e1n\u00edm knihovny CK na report\u00e9rov\u00fdch bun\u011b\u010dn\u00fdch lini\u00edch modeluj\u00edc\u00edch jak p\u0159irozen\u00fd cirkadi\u00e1nn\u00ed syst\u00e9m, tak FASPS mutaci. Bariatric surgery typically results in changes in adipose tissue volume and metabolic function. However, its efficacy differs considerably among the patients. The aim of this thesis is to improve our knowledge on the mechanisms underlying muscle NST, and its control based on comparative study of cold-induced thermogenesis in strains of mice differing in the susceptibility to obesity. The change in environment after birth represents a physiological stimulus for the adaptive response of the entire organism, including the transition from energy metabolism based on glycolysis in the prenatal period to that based on catabolism of lipids from breast milk. The aim of this bachelor&#8217;s&nbsp;thesis is to describe the metabolism of the newborns, including premature babies. The definition of the so-called thermoneutral zone for laboratory animals is not entirely clear and is not well defined for different ages, sexes, genetic backgrounds, and numbers of animals in a cage. Our experimental work is concentrated on\u00a0<a href=\"http:\/\/www.fgu.cas.cz\/en\/articles\/215-the-role-of-synaptic-transmission-modulation-in-pain\">the modulation of nociceptive information at the spinal cord level<\/a>\u00a0that is the first relay center between the periphery and higher brain areas. Our goal is to study these modulatory mechanisms in order to improve therapy for pain conditions such as neuropathic and cancer related pain. This project will be focused on the role of neuroinflammation in modulation of synaptic transmission and chronic pain development. Lately we are interested in the role of TRPV1 receptors, cannabinoids, endogenous lipids, opioids and inflammatory cytokines in this process. In our research we use mainly electrophysiological, optogenetic, OMICS analysis, \u00a0functional imaging, immunohistochemical, molecular and behavioral methods. In collaboration with clinics we aim to study human pathology in pain patients. This project will be focused on the role of neuroinflammation in modulation of synaptic transmission and chronic pain development. Lately we are interested in the role of TRPV1 receptors, cannabinoids, endogenous lipids, opioids and inflammatory cytokines in this process. In our research we use mainly electrophysiological, optogenetic, OMICS analysis, \u00a0functional imaging, immunohistochemical, molecular and behavioral methods. In collaboration with clinics we aim to study human pathology in pain patients. Changes in <em>GRIN<\/em> genes encoding NMDAR subunits are associated with neurodevelopmental disorders. The student will use patch-clamp electrophysiology and live-cell Ca<sup>2+<\/sup> imaging to study NMDAR function, synaptic transmission and plasticity, and circuit properties in neuronal preparations expressing patient <em>GRIN<\/em> gene variants.
