Although the physiological role of NMDAR-modulating sterols and steroids is not well understood, they may influence brain development and cognitive function. We have described a number of synthetic neurosteroid analogues with either inhibitory or potentiating effects on NMDARs. Thanks to this research, neurosteroid NMDAR modulators may become a new class of compounds in the pharmacological toolbox for treating patients with NMDAR dysfunction. <\/p>\n<\/div>\n<p><img fetchpriority=\"high\" decoding=\"async\" class=\" wp-image-36577 aligncenter\" src=\"https:\/\/fgu.cas.cz\/wp-content\/uploads\/2024\/11\/glutamat3-300x106.png\" alt=\"-\" width=\"402\" height=\"142\" title=\"\" srcset=\"https:\/\/fgu.cas.cz\/wp-content\/uploads\/2024\/11\/glutamat3-300x106.png 300w, https:\/\/fgu.cas.cz\/wp-content\/uploads\/2024\/11\/glutamat3-1024x361.png 1024w, https:\/\/fgu.cas.cz\/wp-content\/uploads\/2024\/11\/glutamat3-768x270.png 768w, https:\/\/fgu.cas.cz\/wp-content\/uploads\/2024\/11\/glutamat3.png 1380w\" sizes=\"(max-width: 402px) 100vw, 402px\" \/><\/p>\n<p>Recordings of evoked excitatory postsynaptic currents in hippocampal microisland cultures reveal bidirectional modulation of synaptic NMDA receptors by endogenous neurosteroids: pregnanolone sulfate (PA-S) inhibits NMDA receptors, while pregnenolone sulfate (PE-S) has a potentiating effect.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>The function of NMDARs is regulated by a number of endogenous allosteric modulators, including membrane cholesterol and its metabolites, and endogenous neurosteroids.
